林蛙皮甲醇提取物对小鼠无水乙醇型胃溃疡的保护作用 被引量:1

摘要 [目的]观察林蛙皮甲醇提取物对无水乙醇型小鼠胃溃疡的影响,并探讨其机制.[方法]取昆明种小鼠随机分为模型对照组(生理盐水)、阳性对照组(0.22 g/kg西咪替丁)和林蛙皮甲醇提取物小、中、大剂量组(0.22,0.45,0.90 g/kg).连续给药7 d,末次给药1 h后灌胃给予0.01 mL/g无水乙醇制作胃溃疡模型,30 min后取胃,测定溃疡指数.应用一氧化氮合酶抑制剂N-硝基-L-精氨酸甲酯(L-NAME)观察林蛙皮甲醇提取物的作用机制.[结果]与模型对照组比较,林蛙皮甲醇提取物小、中、大剂量组小鼠溃疡指数均降低,其中高剂量组的溃疡指数显著降低(P<0.01),给予L-NAME后林蛙皮甲醇提取物对小鼠胃溃疡的保护作用受到抑制(P<0.01).[结论]林蛙皮甲醇提取物对小鼠无水乙醇型胃溃疡有保护作用,其机制可能与诱导一氧化氮生成有关. OBJECTIVE To study the protective effects and its mechanism of the methanol extracts of Rana chensinensis skin on ethanol-induced gastric ulcer in mice.METHODSKunming species mice were randomly divided into model group(saline solution,MD),control group(0.22g/kg,CD),and small,middle and large dosage groups of the methanol extracts of Rana chensinensis skin(0.22,0.45,0.90g/kg).All of the mice were administered by gastric gavage for 7th days,and absolute ethanol(0.01mL/g) was given to each mouse in these groups 1 hour later to establish gastric ulcer models,and the mice were sacrificed at 30 min after the administration of ethanol to take out the stomach and the ulcer index(UI) was detected.The mechanism of the methanol extracts of Rana chensinensis skin was observed by using the N-nitro-L-arginine methyl ester(L-NAME).RESULTS The UI was significantly reduced in all of dosages groups of the methanol extracts of Rana chensinensis skin than in model group,especially in large dosage group(P0.01).The protective effects of the methanol extracts of Rana chensinensis skin was partly inhibited by L-NAME(P0.01).CONCLUSIONThe methanol extracts of Rana chensinensis skin has the protective effects for the ethanol-induced gastric ulcer in mice,and its mechanism may be related with NO formation inducted by the methanol extracts of Rana chensinensis skin.
