摘要
为给贵州小香羊品种遗传资源的合理开发利用及进一步选育提供科学依据,同时为更全面地了解其种质特性提供基础资料,利用PCR-RFLP技术对67只贵州小香羊生长激素基因外显子1到内含子2序列、外显子3到外显子5序列分别进行HaeⅢ和FokⅠ内切酶酶切位点多态性检测。结果表明,所扩增的生长激素基因外显子1到内含子2序列存在HaeⅢ酶切位点多态性,并表现为2种基因型AA和AB,基因频率分别为0.73和0.27,等位基因A的频率(0.87)高于等位基因B(0.13);外显子3到外显子5序列中未检测到FokⅠ酶切位点多态性。
To provide scientific basis for the rational development and utilization and further breeding of Guizhou small Xiang goats and basic information for comprehensively understanding its germplasm characterstics,67 Guizhou small Xiang goats were used in the study.PCR-RFLP method was conducted to test the polymorphism of the region between the exon 1 and intron 2,and the region between the exon 3 and exon 5 of GH gene digested with Hae Ⅲ and FokⅠrespectively.The results showed that Hae Ⅲ enzyme polymorphic site was found in GH gene region between the exon 1 and intron 2 by amplification and presented as AA and AB genotype,the frequencies of AA and AB were 0.73 and 0.27,the allele frequency of A(0.87) was higher than that of B(0.13).FokⅠenzyme polymorphic site was not found in the region between the exon 3 and exon 5.
