实验性兔腰椎间盘退变模型中Fas/FasL基因表达及意义 被引量:1

摘要 目的建立腰椎间盘退变模型,检测退变髓核中Fas/FasL基因表达水平,阐明Fas/FasL基因的过度表达是导致髓核细胞凋亡的主要因素之一,腰椎间盘在Fas/FasL基因的过度表达的情况下可能导致超常的凋亡,最终导致椎间盘退变甚或突出。方法选用20只清洁级成年日本大耳白兔,按体重编号,随机分为2组,即模型组和对照组。模型组:麻醉后,手术造成腰椎间不稳;并加以异常应力,以促进造模。对照组(n=10):对照组同法麻醉,作皮肤切口后直接缝合;两组动物在相同条件下饲养,于术后1M、2 M随机各取5只兔子行病理学检查证明造模成功,同时检测两组动物髓核中Fas/FasL基因表达水平。结果 1术后2月模型组兔腰椎间盘2造模2月后,对照组兔腰椎间盘病理学检查无明显退变表现;而模型组造模2个月后,出现纤维环断裂,髓核向后突出,说明造模成功。2检测本实验提取的总RNA纯度高,RT-PCR产物经1.5%琼脂糖凝胶电泳后紫外透射仪下可见扩增之特异产物条带,且目的基因片段产物与引物设计之目的片段大小完全吻合;Fas mRNA、FasL mRNA在退变腰椎间盘组织中有过度表达,与正常腰椎间盘组织相比,其差异有显著性(P<0.05)。结论腰椎间盘退变模型髓核中有Fas/FasL基因表达,其表达水平与正常腰椎间盘组织相比,其差异有显著性(P<0.05);说明Fas/FasL基因的过度表达是导致髓核细胞凋亡的主要因素之一,腰椎间盘在该基因的过度表达的情况下可能导致超常的凋亡,最终导致椎间盘退变甚或突出。
