金银忍冬茎和果实中绿原酸的提取及含量测定 被引量:8

摘要 目的:为忍冬属植物中绿原酸的提取及含量测定寻求更适合的方法。方法:采用超声技术提取金银忍冬茎、果实中的绿原酸,高效液相色谱法测定绿原酸的含量,色谱柱为C-18柱,流动相为甲醇-0.4%磷酸(35:65),流速为1mL/min,检测波长为327nm。结果:仪器精密度良好(RSD=1.41%),测定方法具有较好的重现性试验(RSD=1.56%),供试品溶液在8h内基本稳定(RSD=1.70%)。绿原酸在1.06μg-6.36μg范围内与峰面积具有良好线性关系(R2=0.999 4),平均回收率为98.79%(RSD=1.65%)。金银忍冬茎和果实中绿原酸的含量分别为0.796%,0.379 8%。结论:超声提取时间缩短到15m in,流动相中用甲醇-0.4%磷酸代替了文献中的乙腈-0.4%磷酸,为忍冬属植物中绿原酸的提取及含量测定提供了更为经济、便捷的方法。 Objective: To seek for more suitable methods of extraction and determination of Chlorogenic Acid from Lonicera. Methods : clorogenic acid (CHA) was extracted from stem and fruit of Lonicera maackii with Ultrasonic wave and determined by HPLC on a C - 18 column, methanol - 0.4% H3PO4 (35 : 65) as a mobile phase, 1 mL/min of flow rate, detection wavelength at 360 nm. Results : Good precision ( RSD = 1.41% ) and repeatability ( RSD = 1.56% ) were obtained. The sample was stable within 8 hours ( RSD = 1. 70% ). The linear range of chlorogenic acid was 1.06 - 6. 36μg ( R2 = 0.9994). And the average recovery was 98.79% ( RSD = 1. 65% ). CHA content of chlorogenic acid in stem and fruit of Lonicera maacki were 0. 796% , 0.3?98% respectively. Conclusions : Ultrasonic time was shorten to 15min, the flowing ratio was methanol -0.4% phosphoric acid replaced acetonitrile -0.4% reported in literature. This study offered more economic and convenient method of CHA extraction and determination in Lonicera.
