摘要
目的探讨非小细胞肺癌(NSCLC)组织新型抑癌基因Runt相关转录因子3(RUNX3)和p16的表达及其与肿瘤生物学行为的关系。方法应用免疫组化SP法,对72例NSCLC组织及其癌旁组织RUNX3和P16蛋白表达进行检测。结果 RUNX3和P16的阳性染色均定位于细胞核,NSCLC组织RUNX3阳性表达率为30.56%,P16阳性表达率为41.67%;癌旁组织二者阳性表达率分别为88.89%、93.06%,差异有显著性(χ2=50.925、43.241,P均<0.01)。RUNX3表达强度与NSCLC的组织学分级、临床分期及淋巴结转移有关(χ2=6.244~17.780,P均<0.05),P16表达强度与NSCLC的组织学分级、临床分期、淋巴结转移及原发癌大小有关(χ2=11.530~17.820,P均<0.05)。肺癌组织中RUNX3和P16的表达强度呈正相关(r=0.64,P<0.01)。结论 RUNX3和P16在NSCLC的发生、发展过程中起作用,联合检测RUNX3和P16可作为判定NSCLC恶性程度及评估其侵袭性、转移性的参考指标。
Objective To investigate the expressions of new types of anti-oncogenes RUNX3 and P16 in patients with non-small-cell lung cancer(NSCLC) and their relationship with biological behavior. Methods Immunohistochemical streptavidin-peroxidase method was used to detect the expressions of RUNX3 and P16 protein in 72 NSCLC tissue and their paraneoplastic samples. Results Positive staining of RUNX3 and P16 protein was found in nuclei of NSCLC cells.The positive expression rates of RUNX3 and P16 in NSCLC tissue samples were 30.56% and 41.67%,respectively;and that in paraneoplastic samples were 88.89% and 93.06%(χ2=50.925,43.241;P0.01).The expressive intensity of RUNX3 was correlated with the histological grade,clinical stage and lymph node metastasis of NSCLC(χ2=6.244-17.780,P0.05);and that of P16 in the NSCLC samples was correlated with histological grade,clinical stage,lymph node metastasis and the size of the primary cancer(χ2=11.530-17.820,P0.05).There was a positive correlation between the expressions of RUNX3 and P16(r=0.64,P0.01). Conclusion RUNX3 and P16 play a part in carcinogenesis and progression of NSCLC,a combined detection of RUNX3 and P16 may serve as reference parameters to evaluate the extent of malignancy,invasion and metastasis of this disease.
