摘要
方法:雄性3月龄SD大鼠随机分为对照组(n=20)和长期耐力训练组(n=20),长期耐力训练组从3月龄时开始进行6个月的耐力训练,建立长期耐力训练运动模型。使用免疫组织化学法结合计算机图像处理技术,对各组大鼠心脏血管紧张素Ⅱ的分布和表达进行观察和测定;使用实时荧光定量PCR技术,对各组大鼠心肌Bcl-2和Bax mRNA的表达进行测定;使用酶链反应吸附法测定各组大鼠血清氧化型谷胱甘肽的含量。结果:E组大鼠心脏血管紧张素Ⅱ的表达与C组相比明显上升(P<0.05);E组心肌中Bax mRNA的表达较C组有显著性增高(P<0.05),Bcl-2 mRNA的表达与C组相比没有显著性变化;E组血清氧化型谷胱甘肽的含量显著高于C组(P<0.05)。结论:长期耐力训练后,大鼠心脏血管紧张素Ⅱ表达增强,通过氧化应激途径加剧了心肌细胞凋亡的程度。
Objective: To study the effect of local angiotensin Ⅱ on cardiac apoptosis after long-term endurance training.Methods: Male 3 month-old SD rats were randomly divided into control group(n=12) and long-term endurance training group(n=12).After inducing the animal model by six months-endurance training,using the immunohistochemistry and computerized image system to observe the distribution and expression of local angiotensin Ⅱ in rat heart of each group,using the Real-Time PCR system to analysis the mRNA expression of Bcl-2 and Bax in rat heart of each group,using the Elisa system to analysis the level of serum GSSG of each group.Result: The immunity positive reaction of cardiac local angiotensin Ⅱ of E group increased significantly than C group(P0.05).The mRNA expression of Bax of E group increased significantly than C group(P0.05) and there is not significantly different of the Bcl-2 between E group and C group.The level of serum GSSG of E group increased significantly than C group(P0.05).Conclusion: The find suggests after long-term endurance training,the expression of local angiotensin Ⅱ elevated,and can aggravate cardiac apoptosis by oxidative stress.
