肿瘤组织与正常组织对比切片在病理实验教学中应用

APPLICATION OF NORMAL-AND-TUMOR-TISSUE-CONTRAST SECTION IN TEACHING OF PATHOLOGICAL EXPERIMENT

目的探讨肿瘤组织与正常组织对比切片在病理实验教学中的应用价值。方法分别应用常规染色(HE染色)、特殊染色和免疫组化技术,对50例肿瘤组织及相应的正常组织标本进行染色。结果镜下观察切片正常组织与肿瘤组织3种染色对比明显,HE染色切片可见肿瘤组织细胞的形态、排列与正常组织明显不同,癌细胞胞核大而不规则,核质比例变小;特殊染色可见正常组织与肿瘤组织的弹性纤维、网状纤维及胶原纤维的分布和含量不同;免疫组化染色可见正常组织不或极低表达P53和增殖细胞核抗原(PCNA),CD34阳性标记细胞少、规则,肿瘤组织P53和PCNA表达显著增多,CD34阳性标记细胞多,且分布、排列等不同于正常组织。结论肿瘤组织与正常组织对比切片易于观察、比较和分析,提高了病理学实验教学的效果。

Objective To investigate the value of applying normal-and-tumor-tissue-contrast section in teaching of pathological experiment.Methods By employing conventional staining,special staining and immunohistochemical technique,50 tumor tissue specimens and their corresponding normal tissue were stained.Results Microscopically,three different staining techniques made obvious contrast between normal and cancer tissue.The morphology and arrangement of cancer cells were obviously different from that of normal tissue cells in HE staining,with cancer cells having big irregular nuclei and smaller nuclear-cytoplasmic ratio.The distribution and content of elastic fibers,reticular fibers and collagen fibers were different between normal and cancer tissues with special staining.Immunohistochemical staining showed no or very low expressions of P53 and PCNA in normal tissue,with a small amount of regularly arranged CD34 positive cells were found.P53 and PCNA expressions were markedly increased in cancer tissue,where more CD34 positive cells found with distribution and arrangement different from that of normal tissue.Conclusion Tissue-contrast slice of normal and corresponding tumor tissue is easy to observe,compare and analyze,which improves the teaching result of pathological experiment.
