摘要
目的观察芥菜籽提取物(MSE)对人结肠癌细胞系SW 480细胞凋亡的影响并初步探讨其可能的机制。方法采用体外细胞培养技术,用CCK-8法观察MSE对人大肠癌SW 480细胞生长的影响;Hoechest3325荧光染色显微镜下观察凋亡细胞形态学改变,流式细胞仪检测细胞凋亡;Caspase-3活性检测试剂盒分析其凋亡机制。结果在浓度0.2~1.0 mg/m l作用24~72h范围内,MSE对SW 480细胞增殖具有明显抑制作用(P<0.01)。24h内其凋亡率随着药物浓度的增加而增加,且逐渐从早期凋亡走向晚期凋亡,呈浓度依赖性;24 h内Caspase-3活性较对照组显著提高(P<0.05)。结论 MSE能有效抑制体外培养的人结肠癌细胞SW 480的生长,这种抑制作用与MSE促进肿瘤细胞凋亡、激活凋亡蛋白酶Caspase-3的活性有关。
Objective To observe the effects of mustard seed extract (MSE) on human colorectal cancer SW480 cellular apoptosis, primarily explore its possible mechanism. Methods Take cell culture in vitro. The effects of MSE on cell proliferation were observed by CCK-8 method in vitro. The morphologic changes of apoptosis cell were evaluated by Hoechest33258 fluorescent staining. The cell apoptosis was tested by flow cytometer, and the activity of Caspase-3 was tested by Caspase-3 fluorometric assay kit. Results MSE exhibited a marked anti-proliferative effect on SW480 ceils within the concentration from 0.2 to 1.0 mg/ml and the time from 24 to 72 hours(P 〈0.01 ). The apoptotic ratio was dose-dependant in 24 hours. The activity of Caspase-3 had a significant increase compared to the control group in 24 hours ( P 〈 0.05 ). Conclusions MSE can inh/bit the proliferation of SW480 cell effectively. The effect may be correlated with the tumor cell apoptosis and the Caspase-3 activity induced by MSE
