Notch-RBP-Jκ路径与HPV病毒感染的宫颈病变的相关性研究

摘要 目的观察RBP-Jκ基因在人乳头瘤病毒感染的不同病变宫颈组织中的表达情况。方法采用PCR-DNA反向斑点杂交技术对193例宫颈脱落细胞标本进行HPV基因诊断和分型,RT-PCR方法检测不同病变的宫颈脱落细胞标本的RBP-Jκ基因表达情况;免疫组织化学方法观察RBP-Jκ在不同宫颈组织中表达情况。结果 193例标本中,双重感染36例(18.65%),多重感染4例。高危感染153例(79.2%),其中HPV16感染率最高,为38.86%(75/193)。不同宫颈病变的HPV感染有所不同,随着病变程度的增加,HPV亚型分布有所变化,高危型HPV检出率有所增加。RBP-Jκ/GAPDH基因在慢性宫颈炎组、CIN组、宫颈癌组中的表达分别为0.262±0.233、0.499±0.422、1.094±0.466,3组间的差异有统计学意义(P<0.001)。结论 RBP-Jκ相关的Notch信号通路在宫颈癌细胞中有显著的表达,提示RBP-Jκ的活化与HPV病毒感染后宫颈组织恶变存在一定的相关性。 Objective To investigate the role of RBP-Jκ in different HPV infected cervical cell. Methods All of 193 cases were detected for HPV genotypes by using DNA hybridization technique. The data was analyzed with the statistical software SPSS 17.0. RT-PCR and immunochemistry were employed to assay the expression of Notch pathway signal molecules RBP-Jκ. Results In total 193 cases, 36 (18.7%) diplex infection and four multiple infection. Of positive specimen, 153 (79.2%) were high-risk HPV infection, including 75 (38.9%) HPV 16 type. In different HPV infected cervical cell, as the level of cervical lesions increased, the detection rate of high risk type HPV increased. The expression levels of RBP-Jκ mRNA in cervi- cal lesions of different cervical exfoliated cells were different (P〈0. 001). It was 0. 262±0. 233 in chronic cervicitis group, 0. 499±0. 422 in cervical intraepithelial neoplasia (CIN) group, and 1. 094 ±0. 466 in cervical carcinoma group. Conclusion RBP-Jκ-dependent Notch signaling express specifically in cervical cancer cells and this expression could provide one such mecha- nism that needs to be activated for cervical carcinogenesis.
