摘要
目的 观察高原环境下,兔眼球钝挫伤后视网膜中脑红蛋白(NGB)及角膜中转化生长因子β2(TGF-β2)的表达.方法 将38只兔(76眼)随机分为对照组2只、亚高原组18只(海拔1510m)及高原组18只(海拔3840m).后两组造成兔双眼钝挫伤模型,伤后2 h、6 h、1 d、3 d、7 d、14 d取材切片,行HE及免疫组织染色,病理学彩色图像分析仪测定灰度值,观察视网膜NGB及角膜TGF-β2含量变化.结果 视网膜中NGB在两模型组中的表达均呈现时相性,伤后7 d灰度值最小,高原组与亚高原组比较2 h、6 h、1 d及14 d差异有统计学意义(均P<0.05);伤后角膜中TGF-β2在两模型组中的表达也均呈现时相性,2 h到3 d灰度值逐渐减小,14 d时恢复正常水平,两模型组比较各时间点灰度值差异均有统计学意义(P<0.05);用Pearson相关性分析显示,视网膜NGB与角膜TGF-β2无相关性(r=-0.57,P>0.05).结论 高原环境下,眼球钝挫伤后视网膜NGB及角膜中TGF-β2早期表达增加,并分别呈现时相性,提示NGB和TGF-β2参与缺氧环境下视觉系统组织的损伤与修复,在高原环境眼外伤综合治疗方面有一定的应用前景.
Objective To investigate the Effect of high-altitude(3,840 m) on neuroglobin (NGB)and transforming growth factor-β2 ( TGF-β2 ) expression in rabbit with ocular trauma. Method 38 rabbits (76 eyes) were randomly divided into 3 groups, 2 rabbits in control group (eyeballs without injury), 18rabbits in sub hight-altitude group ( 1510 m) and 18 rabbits in high-altitude group. And ocular trauma mod els were established in the last two groups ( both eyeballs were injuried). At the time point of 2 hours, 6hours, 1 day, 3 days, 7 days, 14 days after injury, the eyeballs were taken out. NGB in the retina and TGF-β2 in the cornea were detected by using histology and immunohistochemical techniques respectively.Results NGB' gray scale value was changed with time in the model groups, 7 day value was the smallest ( P 〈0.05). The differentia of value was statistically significant between the two model groups in 2 hours, 6hours, 1 day, 14 days (P 〈0.05 ). The gray scale value of TGF-β2 in cornea also changed with time in the model groups. From 2 hours to 3 days after injury, the value became smaller and smaller ( P 〈 0.05 ), but on 14 day it reached normal level. The value at each time point was significamtly different between the two medel groups (P 〈 0.05 ). The Pearson correlation method shows that there was no relationship between NGB and TGF-β2 (P 〉0.05 ). Conclusion The expression of NGB and TGF-β2 increased in high-altitude environment after ocular trauma, and it was changed with time respectively. They might take part in the ocular tissue recovery. That indicated their prospects in clinical therapy for ocular trauma.
