范德他尼联合5-氟尿嘧啶对肝癌细胞HepG_2的作用研究

摘要 目的探讨范德他尼(ZD6474)联合5-氟尿嘧啶(5-FU)对肝癌细胞HepG2的杀伤作用。方法用MTT法检测ZD6474、5-氟尿嘧啶单药及其联合对HepG2细胞的增殖抑制,同时采用流式细胞仪检测细胞周期及凋亡情况。结果 ZD6474单药及5-FU单药对HepG2均有明显抑制作用,两药联合具有明显的协同作用(P<0.05)。ZD6474主要使细胞阻滞于G0/G1期,5-FU则使细胞阻滞于S期。两药联合治疗同时产生G0/G1及S期阻滞。联合组凋亡率显著高于任一单药组,差异有统计学意义(P<0.05)。结论 ZD6474和5-FU对肝癌HepG2细胞能产生增殖抑制及促凋亡作用,两药联合表现为协同或相加作用。 Objective To investigate the killing effect of ZD6474 in combination with 5-fluororacil on hepatocarcinoma cells line HepG2.Methods The inhibitory effect of ZD6474 and 5-fluororacil alone and their combination treatment on the proliferation of HepG2 cell in vitro was assessed by MTT assay.The cell cycle and cell apoptosis were detected by flow cytometry.Results ZD6474 and 5-fluororacil alone both significantly inhibited the proliferation of HepG2 cells,showing a synergistic effect of their actions in combined use(P0.05).ZD6474 and 5-fluororacil alone caused cell cycle arrest at G0/G1 and S phases,respectively.Combined use of the two drugs resulted in significant reduction of the S-phase cell percentage and cell cycle arrest at G0/G1 and S phases.The combination use of the two drugs significantly increased the apoptosis rate of the cells as compared with that of the cells with ZD6474 or 5-fluororacil treatment alone(P0.05).Conclusion ZD6474 and 5-fluororacil show a synergistic effect in inhibiting the proliferation and inducing apoptosis of HepG2 cells.
