摘要
目的:建立及评价碱性成纤维细胞生长因子(b F G F)的125Ⅰ标记及放射免疫分析( R I A)方法。方法:采用氯胺 T 氧化法进行 b F G F 125Ⅰ标记,并以双抗体 P E G 的 R I A 测定家兔血清 b F G F水平。结果:125Ⅰ标记b F G F的碘利用率为8198% ,比放射性为303 M Bq/μg,放化纯度达 950%以上;b F G F 放免分析灵敏度可达 04 ng。结论:本研究为 b F G F 的125Ⅰ标记及定量测定提供了简便可靠的方法。
Objective:The present study was to establish and estimate a 125 Ⅰ label and an radioimmunoassay (RIA) of basic fibroblast growth factor (bFGF).Methods:The iodinated recombinant human bFGF (rhbFGF) was prepared using chloramine T (Ch T),and levels of bFGF in rabbit serum were determind by a double antibody polyethylene glycol (PEG) RIA protocol.Results:The radioiodine utilization ratio,specific radioactivity and radiochemical purity of 125 Ⅰ labeling bFGF were 81.98%,3.03 MBq/μg and over 95%,respectively.The sensitivity of bFGF RIA reached 0.4 ng.Conclusion:The present study may provide a convenient and reliable method for 125 Ⅰ label and quantitative determination of bFGF.
