摘要
为了研究ColE2质粒复制起始部位中的一些位点和其生物活性的关系,用人工合成寡核苷酸定点引入点突变的方法,对ColE2质粒复制起始部位的1个位点引入3种点突变,并测定了突变的ColE2质粒复制起始部位的Rep蛋白结合和复制活性。结果表明,该位点的突变导致ColE2质粒复制起始部位的生物活性明显下降。说明该位点对ColE2质粒复制起始部位和Rep蛋白的结合及复制起始都起一定作用。
In order to study the structure and function of the replication origin in plasmid ColE2,mutations were introduced into the position 13 in ColE2 origin.The assays of the in vivo Rep proteinbinding and replication activities of the mutant ColE2 origins show that the mutation of this position leads to a remarkable decrease of the biological activities of ColE2 origin.It is concluded that this position is important to the functions of ColE2 origin.
