人骨髓基质细胞的长期培养及其对单纯疱疹病毒的敏感性

The Longterm Culture of Human Bone Marrow Stromal Cells in Vivo and Their Susceptibility to Herps Simplex Virus

采用静置贴壁培养法，体外长期培养了胎儿、儿童和成人骨髓基质细胞，并将传至５代以上的肌样骨髓基质细胞采用微量细胞病变（ＣＰＥ）法，对单纯疱疹病毒进行了敏感性试验。结果：人骨髓基质肌样细胞对Ⅰ型和Ⅱ型单纯疱疹病毒均敏感，能产生明显的ＣＰＥ，其效价（ＴＣＩＤ５０）可达１０－３～１０－４，其中胎儿骨髓基质肌样细胞对病毒敏感性比儿童高２倍，比成人高４倍。提示此法可为人骨髓细胞体外长期培养提供简便方法，且为该细胞对病毒的敏感性提供实验依据，并为该细胞的利用开辟了新的应用前景。

The fetal child and adult bone marrow stromal cells were cultured for longterm in vitro by the adherent assay in order to investigate their susceptibility to virus.The myoid bone marrow stromal cells were obtained after generating more than five time,then assayed for their susceptibility to Herps simplex virus by microdse of CPE.Results:All myoid stromal cells were susceptible to HSV and .producing obvious CPE with TCID50 up to 10-310-4.Fetal myoid stromal cells were two times higher than child,and four times than adult in terms of virus susceptibility.The research supplies not only a simple practicable assay for exvivo longterm culture of human bone marrow stromal cell but also experimental reference for estimating the virus susceptibility of these cells,at the same time,contributes to a more extensive prospect for applying these cells.
