Caspase-3和Caspase-6在胃癌组织中的表达及意义 被引量:7

摘要 目的检测Caspase-3、Caspase-6基因在胃癌组织、癌旁正常组织中的表达,探讨两者与临床、病理因素的关系及两者之间的关系。方法采用RT-PCR方法检测42例胃癌患者的癌组织、癌旁正常组织中Caspase-3、Caspase-6基因的表达情况,并分析这2个基因与临床、病理因素之间的关系。结果 Caspase-3、Caspase-6基因在胃癌组织中表达率分别为38.09%(16/42)、31.05%(13/42),在癌旁正常组织中的表达率分别为78.57%(33/42)、69.05%(29/42),两者在胃癌组织和癌旁正常组织中的表达差异有统计学意义(P<0.01)。Caspase-3、Caspase-6基因的表达与患者的年龄、性别、肿瘤大小差异无统计学意义(P>0.05),与胃癌肿瘤浸润深度、肿瘤分化程度差异有统计学意义(P<0.05)。Caspase-3基因的表达与淋巴结转移差异有统计学意义(P<0.05),Caspase-6基因的表达与淋巴结转移差异无统计学意义(P>0.05)。在胃癌组织中,两者之间的表达差异无统计学意义(P>0.05)。结论 Caspase-3、Caspase-6基因的表达可能与胃癌的发生、发展相关,对胃癌发生、发展的过程起到下调的作用。联合检测Caspase-3、Caspase-6基因的表达更能反映胃癌的生物学行为,优于单项检测。 Objective To detect the expression of Caspase-3 and Caspase-6 in gastric cancer and adjacent normal tissues,and to explore the relationship among Caspase-3,Caspase-6 and clinicopathologic parameters.Methods The expression of Caspase-3 and Caspase-6 gene was detected by RT-PCR in 42 cases of gastric cancer and 42 cases of adjacent normal tissue,and their relationships with the clinicopathologic parameters of gastric cancer were analyzed.Results There were obvious differences in Caspase-3 expression and Caspase-6 expression between gastric cancer tissues(38.09% and 31.05%,respectively) and adjacent tissues(78.57% and 69.05%,respectively)(P0.01).The expression of Caspase-3 and Caspase-6 was not correlated with the age,gender and tumor size(P0.05),but was correlated with tumor invasion depth and tumor differentiation(P0.05).The expression of Caspase-3(P0.05),but not Caspase-6,was correlated with lymph node metastasis(P0.05).In gastric cancer tissues,there was no correlation between Caspase-3 expression and Caspase-6 expression(P0.05).Conclusion The down-regulation of Caspase-3 and Caspase-6 expression may be associated with the occurrence and development of gastric cancer.Combined detection of Caspase-3 and Caspase-6 expression can significantly reflect the biological behavior of gastric cancer compared with the single detection.
