加味四逆散对实验性肝硬化大鼠防治作用的研究 被引量:5

摘要 目的研究加味四逆散对肝硬化大鼠的防治作用,并对其机制进行初步探讨。方法 Wist-ar雄性大鼠48只随机分为4组,每组12只。除正常组外,模型组、西药组和中药组均给予大鼠皮下注射40%四氯化碳(CCl4)花生油溶液2.5 ml/kg,每周注射2次,连续注射11周。连续注射CCl4的最后2周(即已注射9周后),中药组同时用加味四逆散灌胃治疗;西药组以秋水仙碱灌胃治疗;正常组和模型组则给予等体积生理盐水灌胃;各组连续干预2周后,检测各组大鼠肝功能和肝组织病理改变。结果与模型组比较,中药组血清总蛋白(TP)、血清清蛋白(ALB)含量明显升高,丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)含量明显下降,差异均有统计学意义(P<0.01或P<0.05);肝组织病理改变明显减轻;中药组与西药组比较,TP、ALB含量明显升高,ALT、AST含量明显下降,差异均有统计学意义(P<0.01或P<0.05);肝组织病理改变减轻。结论加味四逆散可显著提高TP、ALB含量,降低ALT和AST含量。加味四逆散能有效地保护肝细胞,恢复肝功能,具有较好的防治实验性大鼠肝硬化的作用。 Objective To investigate the effects of Jiawei Sinisan on rat with liver cirrhosis and probe its mechanism preliminarily.Methods Forty eight male Wistar rats were divided into 4 groups randomly with 12 rats in each group.Except for the normal group,the other 3 groups were injected with 40% CCL4dissolved in peanut oil solution with concentration of 2.5 ml/kg twice weekly for 11 weeks continuously,to construct the model of liver cirrhosis.After the injection of CCL4for 9 weeks,the traditional Chinese medicine(TCM)group was given Jiawei Sinisan,the western group was given autumn narcissus alkali,the normal and model groups were administered physiological saline of the same volume for 2 weeks continuously.Then the liver function and liver tissue pathology were observed.Results Compared with the model group,the total protein(TP)and ALB in the TCM group increased obviously,ALT and AST levels decreased evidently(P0.01 or P0.05),while liver tissue pathology in the TCM group alleviated apparently.When compared to the western medicine group,the TP and ALB in the TCM group increased obviously,ALT and AST decreased evidently(P0.01 or P0.05),whereas liver tissue pathology alleviated markedly.Conclusion Jiawei Sinisan could increase the levels of TP and ALB and decrease the ALT and AST levels.So Jiawei Sinisan could protect liver cells and recover liver function effectively,and has a good effect in protecting and treating rats with experimental liver cirrhosis.
