摘要
目的探讨大肠癌干细胞基因Bmi-1在蛋白和RNA的表达水平及其与临床病理的关系。方法采用SYBR Green实时定量RT-PCR技术检测35例大肠癌组织及相应癌旁和远端正常组织中Bmi-1 mRNA的表达;常规HE染色和免疫组化SP法检测肿瘤组织、癌旁组织、正常黏膜组织中Bmi-1蛋白的表达情况;评价基因的表达与临床病理及分期的关系。结果大肠癌组织Bmi-1蛋白的阳性表达率显著高于瘤旁及正常组织(P<0.05);Bmi-1 mRNA的阳性表达在肿瘤与正常(P=0.086)、肿瘤与瘤旁间(P=0.385)无明显差异。在大肠癌中合并淋巴结转移者Bmi-1 mRNA及Bmi-1蛋白表达水平显著高于未转移者(P<0.05);Duke分期C和D期患者Bmi-1 mRNA表达水平明显升高(P<0.05),癌组织浸润深度越深Bmi-1 mRNA及Bmi-1蛋白表达水平越高。肿瘤分化程度越低Bmi-1蛋白表达强度越强。结论 Bmi-1与大肠癌的发生发展密切相关,检测其表达水平对评估大肠癌患者预后及指导今后治疗有一定意义。
Objective To investigate the expressions of Bmi-1 in colorectal cancer and its correlation with clinical pathological characteristics.Methods The expressions of Bmi-1 mRNA in 35 cases of colorectal cancer,adjacent and distal non-cancerous tissues were quantified by SYBR Green real-time RT-PCR.The expression of Bmi-1 protein and relationship with clinical parameters and the differential expression of genes were detected by HE staining and immunohistochemistry (SP method).Results There was no statistically significant difference among cancer tissues,adjacent and distal non-cancerous tissues,and normal tissues for positive expression of Bmi-1 mRNA(P 0.05).The protein expression was significantly higher in cancer tissues than that in normal tissues (P 0.05).The expression of tissue Bmi-1 mRNA in colorectal cancer patients with lymph-node metastasis was significantly higher than those without metastasis.While in patients with deeper infiltration and Dukes'C and D,the expression of Bmi-1 increased significantly.Conclusions Bmi-1 expressions in colorectal cancer are closely related to the infiltration and metastasis of colorectal cancer,which is helpful to evaluate the prognosis and guide the future treatment of patients with colorectal cancer.
