白细胞介素3受体系统及转录因子与急性髓系白血病关系的研究进展

摘要 造血干细胞的自我更新以及定向分化能力,受多种造血生长因子的调控,白细胞介素(IL)3、粒细胞-巨噬细胞集落刺激因子、IL-5在其中发挥重要的作用,这些细胞因子与靶细胞表面受体结合通过激活下游信号通路发挥生物学功能。研究表明急性髓系白血病患者中存在IL-3、粒细胞-巨噬细胞集落刺激因子及其受体的异常表达且常伴有髓系相关的转录因子的突变与功能失调。因此,研究者推测急性髓系白血病分化受阻可能与IL-3受体系统的异常表达有关,而后者又受髓系相关的转录因子的表达调控。 The self-renewal and directed differentiation of hematopoietic stem cell is regulated by a variety of hematopoietic growth factors,such as interleukin(IL)-3,granulocyte-macrophage colony stimulating factor(GM-CSF),IL-5,which played important rolees in the regulation.These cytokines binding with their receptors on the target cells to transduce survival signals via activating the downstream signals.Studies have shown that the abnormal expression of IL-3,GM-CSF and their receptors and the mutations and dysfunction of the transcription factors in human acute myeloid leukemia.Therefore,researchists hypothesized that the blocked differentiation of acute myeloid leukemia may be related to the abnormal expression of IL-3R system and transcription factors.
