摘要
雷公藤甲素(TPL)是从雷公藤植物中提取的环氧二萜类化合物。TPL对胰腺癌体外培养细胞及体内实体瘤细胞均有明显的生长抑制作用,主要通过诱导细胞凋亡或促进细胞自溶这两种不同方式杀伤各种类型的胰腺癌细胞。TPL诱导胰腺癌细胞凋亡的主要作用机制有下调部分原癌基因,抑制热休克蛋白70,活化caspase以及抑制核因子κB的激活。此外,TPL还能增强肿瘤细胞对放化疗的敏感性。
Triptolide(TPL) is a diterpenoid originally isolated from Tripterygium Hook wilfordii.It has been reported that triptolide inhibits the growth of pancreatic cancer cells in vitro and suppresses tumor growth in vivo.TPL kills diverse types of pancreatic cancer cells by inducing apoptosis or promoting cell autolysis.The mechanisms by which triptolide induces apoptosis of pancreatic cancer cells include the down-regulation of some oncogenes,inhibition of heat shock protein 70,activation of caspase,and inhibition of NF-kB activation.Moreover,triptolide sensitizes pancreatic cancer cells to chemotherapeutic agents and ionizing radiation(IR).
