摘要
目的:观察补髓生血颗粒对CAA患者骨髓单个核细胞中AKT磷酸化水平变化,探讨补髓生血颗粒治疗CAA的作用机制。方法:将30例CAA患者予以补髓生血颗粒治疗6个月,另取28例健康志愿者作为正常对照组。采用蛋白质印迹方法(Western-blot)观察CAA患者治疗前后骨髓单个核细胞中pAKT-Ty308的表达水平。结果:CAA患者治疗前骨髓单个核细胞pAKT-Ty308的表达低于正常对照组(P<0.05),经补髓生血颗粒治疗后,pAKT-Ty308的表达有所上升,与治疗前比较有显著性差异(P<0.05)。结论:CAA患者骨髓单个核细胞PAKT-Ty308的水平均呈低水平表达,提示pAKT与CAA发病密切相关,补髓生血颗粒可能通过调节PAKT-Ty308的表达来改善CAA骨髓造血微环境。
Objective: To observe the effect of Busuishengxue Granule on the level of Phosphorylated AKT in CAA patients' bone marrow,and to explore the mechanism of Busuishengxue Granule on CAA.Methods: 30 CAA patients were treated with Busuishengxue Granule for 6 months,and another 10 healthy volunteers as normal control group.Use Western blot method(Western-blot) to observe the expression of PAKT-Ty308 in CAA patients' bone marrow mononuclear cells before and after treatment.Results: The expression of PAKT-Ty308 in CAA patients' bone marrow mononuclear cells were lower than that of normal control group(P0.05),and the expression of PAKT-Ty308 was on the rise after treatment,so the comparison was significant difference before treatment.Conclusion: The expression of PAKT-Ty308 in CAA patients' bone marrow takes on low levels,so it suggests that PAKT is closely related to the pathogenesis of CAA,and Busuishengxue Granule may regulate the expression of PAKT-Ty308 to improve the CAA bone marrow microenvironment.
