摘要
目的:观察钙通道阻滞剂对增生性瘢痕成纤维细胞三维培养模型收缩情况的影响,探讨进一步应用于临床治疗增生性瘢痕挛缩的可能性. 方法: 对人增生性瘢痕成纤维细胞进行体外培养,并构建含成纤维细胞的胶原网络支架(fibroblastpopulated collagen lattice, F P C L),即三维培养系统. AIM:In order to evaluate the possibility of using the calcium antagonist to treat hypertrophic scar contracture in clinically, we investigated the effect of calcium antagonist on the hypertrophic scar derived fibroblast populated collagen lattice contraction. METHODS: Having established fibroblast populated collagen lattice (FPCL) by culturing human hypertrophic scar fibroblast in vitro , we studied the effect of calcium antagonist on FPCL contraction. RESULTS: Six strains of fibroblast were isolated from human hypertrophic scar obtained from operation. Calcium antagonist, verapamil, could inhibit the contraction of FPCL by a does dependent way, but had no effect on the rapid contraction of FPCL. Morphological changes had also been observed. CONCLUSION: Calcium antagonist, verapamil, can inhibit the contraction of hypertrophic scar fibrolast populated collagen lattice. Calcium antagonist may play a vital role in the inhibition, which inhibits not only the changes of Ca 2+ concentration of the cell but also the collagen synthesis.
