The establishment of cleft palate model and observation of its morphogenesis in BALB/C mice

摘要 目的:研究维甲酸和地塞米松联合用药对BALB/C近交系小鼠腭部发育的影响。方法:将BALB/C系孕鼠分为给药组和对照组,在妊娠第12d分别给药,并于妊娠期第138,1320,148,1420,158,1520,168d处死孕鼠,取胎鼠头部做冠状切片,HE染色,进行观察。结果:维甲酸和地塞米松联合用药可诱导BALB/C系小鼠形成腭裂,其主要原因是腭突上抬延迟。结论:维甲酸和地塞米松联合应用诱导BALB/C近交系小鼠形成腭裂是一种稳定的腭裂动物模型。 Objective: To study the effects of retinoic acid(RA) and dexamethasone (DEX) on the palate development in BALB/C mice. Method: Pregnant BALB/C mice were divided into treated group and control group.In the treated group,the mice were dosed on GD12 with RA(100 mg/kg) and DEX(10 mg/kg).The mice were killed on day 138,1220,148,1420,158,1520,168,respectively.The coronal sections of the heads of the embryonic mice were prepared,stained with HE and observed microscopically. Result: Cleft palate was induced in BALB/C mice by the coadministration of RA with DEX,because the development of palate shelves was delayed. Conclusion: Coadministion of RA with DEX can induce cleft palate in BALB/C mice,and can be used to establish stable model of cleft palate.
