摘要
目的观察山姜素对新生大鼠心肌细胞凋亡的影响并探讨其机制。方法利用体外培养的新生大鼠心肌细胞,给予200 mmol.L-1H2O2作用的同时给予不同浓度的山姜素作用24 h,MTT法测定细胞存活率,流式细胞仪分析细胞周期,Annexin V-FITC/PI双标记法测定细胞凋亡率,Western blot检测Caspase-3及Bcl-2表达。结果山姜素可显著抑制心肌细胞凋亡(P<0.01),其作用具有时间和浓度依赖性;同时改变细胞周期分布,与模型组比较,山姜素不同浓度组的细胞凋亡率有显著性差异(P<0.01)。结论山姜素可抑制H2O2导致的心肌细胞凋亡,并且改变细胞周期分布,从而促进心肌细胞的增殖。
Objective To observe the alpinetin in neonatal rat cardiomyocyte apoptosis and its mechanism.MethodsThe neonatal rat cardiac myocytes cultured in vitro give 200 mmol·L-1H2O2 effect as well as the effect to alpinetin with different concentrations 24 h.MTT cell viability was determined by flow cytometry cell cycle analysis;apoptosis rate was detected by Annexin V-FITC/PI double labeling method;Caspase-3 and Bcl-2 expressions were detected by Western blot.Results Alpinetin factors could significantly inhibit cardiomyocyte apoptosis(P0.01) yet its role was dependent on time and concentration,it would change cell cycle distribution,compared with the model group,the apoptosis rate of alpinetin with different concentrations was dramatically different(P0.01).Conclusion Alpinetin can inhibit myocardial apoptosis induced by H2O2 and change the cell cycle distribution,thereby promoting cardiac cell proliferation.
