姜黄素对糖尿病大鼠肾组织NF-κB的DNA结合活性升高的抑制作用 被引量:6

Inhibition effect of curcumin on NF-κB DNA-binding activity increased in the kidney of experimental diabetic rats

摘要 目的探讨姜黄素对糖尿病大鼠肾组织核因子-κB(nuclear factor-κB,NF-κB)的DNA结合活性升高的抑制作用。方法将大鼠随机分为正常对照组(A组)、糖尿病组(B组)及姜黄素治疗组(C组),采用腹腔单剂量注射链脲佐菌素(streptozotocin,STZ)65mg/kg建立糖尿病大鼠模型。治疗组给予姜黄素30mg·kg-1·d-1腹腔注射。实验第4周各组分别宰杀5只大鼠,取部分右肾皮质,采用电泳迁移率改变分析(electro-phoresis mobility shift assay,EMSA)检测姜黄素对糖尿病大鼠肾组织NF-κB的DNA结合活性升高的影响,并观察肾组织的病理变化。结果糖尿病大鼠肾组织NF-κB的DNA结合活性明显升高,姜黄素对其有显著抑制作用,并改善糖尿病大鼠肾脏病变。结论姜黄素通过抑制糖尿病大鼠肾组织NF-κB的DNA结合活性升高而减轻糖尿病大鼠肾脏病变。 Objective To study the inhibition effect of curcumin on DNA-binding activity increased of NF-κB in the kidney of experimental diabetic rats.Method Rats were randomly divided into normal control group(group A),diabetic group(group B),and treatment group(group C).Diabetes models were induced by streptozocin(STZ 65mg/kg) intraperitoneal injection,treatment group was given curcumin intraperitoneal injection(30mg·kg-1·d-1).Five rats were killed in each group at the 4th week,took the renal cortex of right kidney,and detected DNA-binding activity of NF-κB by electrophoresis mobility shift assay(EMSA),and observed the pathological changes of kidney.Results The DNA-binding activity of NF-κB in diabetic group was significantly increased,and curcumin can significant inhibition it.Accordingly,renal pathologic changes were significantly improved.Conclusions Curcumin can decrease the damage of diabetic rats' kidney through inhibiting DNA-binding activity of NF-κB.
