间歇性缺氧/复氧对结肠癌SW480细胞晚期氧化蛋白产物表达的影响

摘要 目的:研究间歇性缺氧/复氧对体外培养SW480细胞生长形态及晚期氧化蛋白产物(AOPP)表达的影响,探讨结肠癌SW480细胞间歇性缺氧/复氧的氧化应激现象。方法:将SW480细胞体外培养48 h后传代,随机分为四组:A组持续缺氧15 min,B组间歇性缺氧/复氧120 min,C组持续缺氧60 min,D组为常氧组,检测细胞培养液中的AOPP含量。结果:A、D组细胞形态无明显改变,B、C组细胞两端突起明显,A、B组和B、C组间AOPP差异显著(P<0.05),B、D组间AOPP存在非常显著差异(P<0.01)。结论:相同缺氧方法、不同缺氧时间SW480细胞生长形态发生改变,且间歇性缺氧/复氧使SW480细胞中的AOPP含量明显升高,发生明显的氧化应激。 Objective: To study the effects of intermittent hypoxia/ reoxygenation on SW480 cell lines culturing in vitro grown appearance and the expression of advanced oxidation protein products (AOPP)and investigate the oxidative stress about the colon cancer SW480 cell lines which intermitted hypoxia/ reoxygenation. Methods: SW480 cells harvested incubated from 48 hours cuhuring in vitro were divided into four groups: group A: continuous hypoxia for 15 minutes, group B: intermittent hypoxia/reoxygenation for 120 minutes, group C: continuous hypoxia for 60 minutes, group D: normoxia group. AOPP was tested in the cell culture fluid. Results: The appearance of group B and C change obviously, compared with grnup A and D,P〈0.05. And it showed the AOPP increased significantly in group B,P〈0.01. Conclusions: Same hypoxia manner but different hypoxia time the grown appearance of SW480 cell lines changed.And intermitted hypoxia/ reoxygenation can certainly increase the expression of AOPP.
