摘要 目的研究肠道病毒71型(EV71)在Vero细胞上传代适应的可行性,为用Vero细胞制备EV71疫苗提供科学依据。方法将EV71 H6株病毒在Vero细胞上进行3次蚀斑筛选并连续传代培养,然后检测适应株的生物学性状及稳定性。结果经筛选后的Vero细胞适应株可被特异抗EV71血清中和。传至第5代,病毒滴度可达8.13 LogCCID50/ml。制备毒种的适宜感染剂量(MOI)为0.01,在96 h左右收获的病毒滴度较高。理化学试验结果表明,EV71核酸类型为RNA,不具有耐热性,但可耐受乙醚、酸的作用。结论 EV71 H6株能较好的适应Vero细胞,并保持稳定的生物学性状。 Objective To study the feasibility of adapting enterovirus 71(EV71) in Vero cells for providing reference of the EV71 vaccine production.Methods EV71 H6 strain was inoculated to Vero cells for plaque screening and further subculture.The biological characters and stability of adapted strain were tested.Results The adaptive strain was neutralized with EV71 antiserum.After EV71 H6 strain was subcultured in Vero cells for 5 passages,the virus titer increased to 8.13 LogCCID50/ml.The optimal multiplicity of infection(MOI) of adapted strain was 0.01,and the optimal harvest time was 96 h.Physicochemically,EV71 was a RNA virus,and sensitive to heat,resistant to aether and acid.Conclusion EV71 H6 strain can be well adapted in Vero cells cultures with stable biological character at the same time.
