Primary location of target antigen in the new passive membranous nephropathy model

摘要 目的探讨兔抗人足细胞蛋白抗体诱导的新型膜性肾病大鼠模型靶抗原的初步定位。方法观察第28天模型组大鼠血清和免疫后的兔血清分别在正常大鼠肾组织、正常人肾组织和正常人足细胞的定位。结果免疫后的兔血清和第28天模型组鼠血清均可见沉积于正常人肾组织、正常大鼠肾组织基底膜外侧和足细胞结合。兔血清和人足细胞的细胞浆和细胞膜均能结合,鼠血清和人足细胞在足突部位结合最多。结论新型被动性膜性肾病模型靶抗原初步定位为足突。 Objective To observe the probable location of the target antigen in the new passive membranous nephropathy model.Methods The rat model was induced by intravenous injection of rabbit anti-human podocyte-protein antiserum.The probable location of target protein was observed in the immunofluorescence by the serum of the rat on the 28th day after antiserum injection and the serum of the rabbit after immunization,which were respectively incubated with the normal rat kidney,the normal human kidney and the normal podocyte after ten days differentiation.Results The immunized rabbit antiserum and the rat serum on the 28th day after antiserum injection respectively incubated with the normal human kidney and the normal rat kidney showed the deposition outside glomerular basement membrane.The rabbit serum incubated with normal podocytes after ten days differentiation showed the deposition in cytoplasm and cell membrane.The rat serum incubated with the podocytes showed the most deposition in the foot process.Conclusion Probable location of target antigen in the new passive membranous nephropathy model is foot process.
