摘要
目的探讨Culiin 1和p53在子宫浆液性腺癌中的表达及临床病理意义,以提高对该病的认识。方法收集10例早期子宫浆液性腺癌病例,并以10例早期高级别子宫内膜样腺癌和10例正常增生期子宫内膜标本作对照,通过HE切片、光镜观察和免疫组织化学方法检测Culiin 1和p53的蛋白表达水平,并用实时RT-PCR方法检测Culiin 1的mRNA水平,结合临床随访资料进行临床病理分析。结果 Culiin 1蛋白和Culiin 1 mRNA在子宫浆液性腺癌和高级别子宫内膜样腺癌中的表达比正常增生期子宫内膜中的明显升高(P<0.01)。p53蛋白在子宫浆液性腺癌和子宫内膜样腺癌呈高表达,而正常增生期子宫内膜阴性。结论子宫浆液性腺癌和高级别子宫内膜样腺癌中Culiin 1蛋白和Culiin 1 mRNA表达增加,伴随p53蛋白表达增加。以Culiin 1为中心的SCF复合体介导的泛素-蛋白酶水解过程失活,可能与上述改变有关。
Objective To research on the expression of Cullin 1 and p53 in endometrial serous adenocarcinoma (ESA) so as to explore the significance of these alterations in this disease. Methods 10 cases of ESA patients (,Stage I and II) were collected, and the expression of Cullin 1 and p53 were determined immunochemically,and the level of Cullin 1 mRNA was examined by real-time ER-PCR.10 caese of high grade endometrial carcinomas (HGEC) with same stages and 10 normal profirative endometium(EM) examples were checked up on the same time for control. Results The expression of Cullin 1 protein and Culiin 1 mRNA in SEAs and HGECs was higher than EMs significantly(P〈0.01);and there was an over-expression of p53 in SEAs and HGECs while negative in EMs. Conclusion There was an increased expression of p53 protein in ESAs and HGECs with an elevation of Cullin 1 protein and Culiin 1 mRNA. The SCF complex,which was centered by Cullin-l,might take part in the above changes.
