摘要
目的探讨丹栀逍遥散对糖尿病抑郁大鼠肝组织胰岛素受体和胰岛素底物-1 mRNA影响的分子机制。方法以高脂高糖乳剂加链尿佐菌素复制糖尿病大鼠模型,再予慢性孤养加慢性不可预见性中等刺激,建立糖尿病抑郁模型,给予丹栀逍遥散3 w后,观察其体重、行为学、空腹血糖、胰岛素、葡萄糖耐量试验、下丘脑-垂体-肾上腺轴(HPA)的激素的变化、采用RT-PCR检测肝组织胰岛素受体(IR)、胰岛素受体底物-1(IRS-1)mRNA的表达。结果丹栀逍遥散提高糖尿病抑郁大鼠活动度;降低血糖和胰岛素;降低CRH、ACTH、CorT;提高胰岛素靶组织肝的IR、IRS-1mRNA的表达。结论丹栀逍遥散治疗糖尿病抑郁其机制可能与纠正紊乱HPA轴和提高肝组织IR、IRS-1的基因表达从而改善胰岛素的信号传导有关。
Objective To observe the effects of Danzhi xiaoyao decoction(DZXYD) on insulin receptor and insulin receptor substrate-1 mRNA expression in liver tissues of diabetes mellitus(DM) rat with depression and investigate the molecular mechanism.Methods Male SD rats were administrated with enriched diet and injected with Streptozotocin(STZ,30 mg/kg),then isolated and independent chromic unpredictable mild stimulation(3 w) to build the model of 2 type diabetes mellitus(T2DM) rat with depression.The changes of behaviors and body weight were observed and the concentration of serum fasting blood glucose(FBG),serum fasting insulin(FINS),oral glucose tolerance test(OGTT),hypothalamus-pituitary-adrenal(HPA) axis function were measured and the expressions of IR and IRS-1 mRNA in liver tissue were determined with RT-PCR.Results Compared with the model group,the body weight and verbal behaviors were increased,FBG,FINS and OGTT levels were decreased significantly,ACTH,CorT level were lower and IR and IRS-1 mRNA expressions were increased in DZXYD group.Conclusions The effect of DZXYD on DM with depression is probably related with adjusting the disorder of HPA axis and increasing IR and IRS-1 mRNA expressions in liver tissue.
