摘要
目的:研究肝硬化大鼠肝脏、脾脏血红素氧化酶(HO-1)、HO-2的定位表达的改变。方法:制备大鼠肝硬化模型,用免疫组化染色法观察HO-1、HO-2在细胞内的分布,并在显微镜下用计算机图像分析技术计算两者表达的强弱。结果:与对照组相比,肝硬化组肝脏、脾脏、门静脉HO-1阳性细胞的表达显著增加,且主要分布在肝脏的门管区周围及脾脏的红髓区,而HO-2的变化不明显。结论:HO可能参与肝硬化的发生过程、与门脉高压血流动力学紊乱有关。
Objective:To explore the expression and distribution of heme oxygenase isoforms(HO-1 and HO-2) in liver and spleen of cirrhotic rats.Methods:Liver cirrhosis was induced by the subcutaneous administration of CCL4.Distributions of HO-1 and HO-2 in rat tissues were examined immunohistochemically using polyclonalantibodies.The pathologic changes and expression of HO-1 and HO-2 in rat tissues were observed by light microscope and compared using image analysis system.Results:Compared with the controls,HO-1 positive cells were more abundant and intensive in liver and spleen of cirrhotic rats and mainly located in portal areas of liver and in red pulp of spleen.However,no statistical difference of HO-2 between cirrhotic rats and controls was observed.Conclusion:The HO-CO pathway may be involved in the development of liver cirrhosis and pathogenesis of the portal hypertension.
