摘要 目的:观察雷公藤总生物碱(Alkaloids of Tripterygium Wilfordii,ATW)的急毒及抗炎镇痛作用。方法:根据半数致死量的方法进行雷公藤总生物碱的急性毒性实验;采用二甲苯小鼠耳肿胀模型、大鼠棉球肉芽肿、小鼠毛细血管通透性实验法研究ATW的抗炎作用;采用小鼠温浴法、超声波甩尾法、热板法和扭体法研究ATW的镇痛作用。结果:雷公藤总生物碱的口服LD50(半数致死量)为5.165 g/kg,腹腔LD50为0.7238 g/kg。ATW能明显抑制小鼠耳廓肿胀,抑制棉球肉芽肿的增重,降低小鼠腹腔毛细血管通透性;ATW对冰醋酸、超声波、热刺激所诱发的小鼠疼痛均有抑制作用。结论:雷公藤总生物碱具有明显的抗炎镇痛作用,但同时有一定的毒性。 Objective: To study Anti-inflammatory,analgesic effect and acute toxicity of alkaloids of Tripterygium wilfordii(ATM).Methods: The acute toxicity was evaluated by median lethal dose(LD 50).Using three animal models of swelling ear,swelling cotton-ball granulation and capillary permeability to determine ATW’s anti-inflammatory action;The analgesic activity of ATW was observed by four animal models,such as warm bath,tail-flick,hot plate and stretching.Results: The LD 50 of ATW was 5.165g/kg when taken orally and 0.7501g/kg when administrated peritoneal injection.ATW evidently inhibited the swelling of mice ear and cotton-ball granulation of rats,reduced the capillary permeability in peritoneal cavity;ATW had analgesic effect on mice pain induced by glacial acetic acid,ultrasonic wave and thermal stimulus.Conclusion: ATW had remarkable Anti-inflammatory and analgesic effect,but it also had some toxicity.
