摘要
目的:探讨蒿鳖养阴软坚方药物血清对肝星状细胞(HSC-T6)的作用。方法:MTT法分析不同剂量的蒿鳖养阴软坚方药物血清对HSC增殖的影响,并测定培养上清中Hyp含量和LDH活力,流式细胞仪检测其对HSC细胞周期的影响。结果:与正常血清组比较,蒿鳖养阴软坚方(0.82 g/kg、2.59 g/kg、8.2 g/kg)药物血清组吸光度(A)值显著降低,抑制率分别为11.26%、30.24%、44.51%;G0/G1期细胞比例增多而S期细胞比例减少;Hyp生成减少,LDH略微升高。结论:蒿鳖养阴软坚方药物血清可能通过阻抑HSC于G0/G1关卡而抑制HSC-T6增殖,同时抑制了胶原蛋白产生,且无细胞毒作用。
Objective: To explore the effects of drug-sera with Haobieyangyinruanjian prescription on hepatic stellate cells(HSC-T6).Methods: MTT colorimetric was used to detect the effect of drug-sera of different doses of HBYYRJ prescription on HSC-T6 proliferation;the contents of hydroxyproline(Hyp) and the activity of lactic dehydrogenase(LDH) in HSC-T6 supernatant were also assayed;HSC-T6 cell cycle was detected with flow cytometry.Results: Compared with normal control serum group,drug-serum groups of HBYYRJ prescription(0.82 g/kg、2.59 g/kg、8.2 g/kg) absorbance(A) value reduced significantly(P〈0.05,P〈0.01),inhibition ratios were 11.26%,30.24%,44.51% respectively;the ratio of HSC at G0/G1 phase increased while the ratio of HSC at S phase decreased;Hyp reduced(P〈0.05,P〈0.01),LDH increased slightly(P〉0.05).Conclusion: Drug-sera of HBYYRJ prescription can inhibit HSC proliferation by blocking HSC at G0/G1 phase from entering DNA synthetic phase(S phase),inhibit collagen generation,without cytotoxic effects.
