摘要 转化生长因子-β(transforming growth factor-β,TGF-β)是一个多功能的生长因子,对角膜上皮、内皮和基质细胞均有影响。TGF-β与免疫抑制有着密切的联系,当在培养液内添加TGF-β1时,可使CD69+CD25+和CD25+CD4+、CD25+CD8+、CD71+CD4+、CD71+CD8+的表达受到抑制。有实验证明:局部应用TGF-β1对大鼠的移植排斥反应具有抑制作用,TGF-β1治疗组移植片比对照组透明时间长。用猪角膜作载体,体外构建TGF-β1转基因后板层角膜植片移植给兔,结果表明:角膜内皮细胞内高表达TGF-β1,可抑制免疫排斥反应的发生和延长移植片的成活时间。初步应用TGF-β1滴眼液治疗角膜与免疫相关疾病结果证实,TGF-β1能够提高这些眼病的治愈率,缩短其病程。 Transforming growth factor-β(TGF-β) is a multifunctional growth factor,which exerts biological functions on corneal epithelial cells,endothelial cells and stromal cells.TGF-β has close relationship with immunosuppression.The expression of CD69+CD25+,CD25+CD4+,CD25+CD8+,CD71+CD4+ and CD71+CD8+ was significantly decreased with treatment of TGF-β1.It has been proved by experiments that local application of TGF-β1 can inhibit the graft rejection and obtain a longer duration of transparent corneal graft in rats.Lamellar corneal graft from pig transfected with TGF-β1 is transplanted in rabbit and the results show high expression of TGF-β1 in corneal endothelial cells,suppression of immunological rejection and increased duration of graft survival.Preliminary application on the treatment of immuno-related keratopathy indicates that TGF-β1 can increase the cure rate and significantly shorten the course of these diseases.
