摘要
目的:探讨维甲酸受体RARα与RXRα在卵巢癌组织中的表达及其与临床病理参数和预后的相关性。方法:应用免疫组织化学方法测定93例原发性卵巢癌组织标本中RARα与RXRα的表达,采用χ2检验分析RARα和RXRα表达与临床病理参数的关系,运用非参数Spearman相关系数检验两者之间的相关性,并对随访结果进行生存分析。结果:93例卵巢癌组织中RARα和RXRα阳性表达率分别为68.8%和52.7%。RARα表达与卵巢癌患者年龄、肿瘤大小及转移(包括淋巴结)无关(P>0.05),而与病理分级、组织学类型及FIGO分期有关(P<0.05)。RXRα表达与卵巢癌患者年龄、肿瘤大小、组织学类型、FIGO分期及转移无关(P>0.05),而与病理分级有关(P<0.05)。经Spearman相关系数检验,RARα与RXRα在卵巢癌中的表达呈正相关(r=0.431,P=0.000),两者协同表达。Cox单因素分析、多因素分析及Kaplan-Meier分析均显示,卵巢癌患者的预后与FIGO分期和RARα表达有关,两者是影响卵巢癌预后的因素(P<0.05),而患者的年龄、肿瘤大小、组织学类型、病理分级及RXRα表达与预后无关(P>0.05)。结论:RARα和RXRα高表达提示肿瘤恶性程度高,FIGO分期和RARα表达与卵巢癌患者的预后关系密切,RARα可作为临床诊断和评估卵巢癌患者预后有价值的生物学指标之一。
OBJECTIVE:To investigate the expressions of retinoic acid receptor RARα and RXRα in ovarian cancer tissues and their correlation with clinicopathological features and prognosis.METHODS: Immunohistochemical staining was used to detect the expressions of RARα and RXRα proteins in 93 primary ovarian cancer tissue samples,and the expressions of RXRα and RARα with clinical pathological characteristic.Results were analyzed by pearson χ2 test and Spearman rank correlation.These 93 cases were followed-up and analyzed their prognosis.RESULTS: In the 93 cases,the postive rates of RARα and RXRα were 68.8% and 52.7%,respectively.There were significant relationship of RARα protein expression with pathological grade,histologic type and FIGO stage(P0.05),but RARα protein expression was unrelated to age,tumor size and metastasis including lymph nodes(P0.05).There were significant relationship of RXRα protein expression with pathological grade(P0.05),but RXRα protein expression was unrelated to age,tumor size,histological type,FIGO stage and metastasis(P0.05).Spearman rank correlation showed a significant correlation between RARα and RXRα(r=0.431,P=0.000).Univariate analysis,multivariate analysis and Kaplan-Meier analysis showed that FIGO stage and RARα expression were related to prognosis(P0.05).Both of FIGO stage and RARα expression retained significance in prospecting prognosis of ovarian cancer.There were no significant relationship of age,tumor size,histological type,histological grade and RXRα expression to prognosis(P0.05).CONCLUSIONS: High expression of RARα and RXRα were suggested highly malignant tumor.FIGO stage and RARα were closed to prognosis.RARα expression may be used as an useful biological marker for diagnosis and prognosis of ovarian cancer.
