口腔黏膜异种移植模型巨噬细胞浸润及他克莫司干预的实验研究

摘要 目的:观察大鼠口腔黏膜异种移植模型急性排斥反应过程中巨噬细胞浸润记数变化及他克莫司的干预作用。方法:Wistar大鼠接受BALB/c小鼠舌体移植建立口腔黏膜异种移植模型,免疫组化法观察颊黏膜急性排斥反应过程中3个时点巨噬细胞记数变化情况。结果:大鼠颊黏膜术后即出现急性排斥反应并随时间推移排斥反应减弱,移植区巨噬细胞浸润程度随病程呈递减趋势。术后第7天移植组巨噬细胞记数为580.00个/HPF,移植用药组325.80个/HPF,术后第14天移植用药组246.45个/HPF,移植组220.00个/HPF,术后第28天移植组173.00个/HPF,移植用药组73.00个/HPF。不同处理组术后3个时点记数差别均有统计学意义(P<0.05),移植组术后3个时点记数差别有统计学意义(P<0.05),移植组与移植用药组3个时点两两比较差别无统计学意义(P>0.05)。结论:成功建立大鼠口腔黏膜异种移植模型,巨噬细胞浸润程度与急性排斥反应病程有关,他克莫司对于巨噬细胞浸润无明显抑制作用。 Objective: To observe the dynamic process and extent of the macrophage infiltration druing acute rejection of oral mucosal xenotransplantation in rats and the therapeutic effect of tacrolimus(FK 506).Methods: Oral mucosal xenotransplantation model was established in Wistar rats accepting the body of BALB/c mouse tongue.In xenotransplantation group(X group) rats received no medication,in xenotransplantation plus medication group(FK506 group) each rat was given tacrolimus at 2 mg/(kg·d) 3 d before operation and at 0.5 mg/(kg·d) after operation.The rats with sham operation were used as the controls.Immunohistochemical assay was performed to detect the counts of the infiltrating macrophage 7,14 and 28 days after operation respectively.Results: Acute rejection occured in the rat oral mucosa after xenotransplation operation immediately,and the reject reaction become weakened as time lapse,macrophage infiltrating extent presented a gradual decrease trend concomitancing acute rejection.The macrophage counts in X group was 580.00/HPF 7 days post operation,in FK506 group was 325.80/HPF;at 14 days post operation that was 220.00/HPF in X group and 246.45 in FK506 group;at 28 days post operation in X group was 173.00/HPF,in FK506 group was 73.00/HPF.There was statistical significance of the macrophages counts in each different handling groups at the time points of 7,14 and 28 days after operation(P0.05).There was no statistical significance between X group and FK506 group(P0.05).Conclusion: The rat oral mucosal xenotransplantation model can be established,macrophage infiltrating extent is related to the course of acute rejection,meanwhile tacrolimus has no obviously depressant effect on the macrophage infiltration.
