摘要
目的研究短期、高剂量全氟辛烷磺酸(pefluorooetanesulfonate,PFOS)经口灌胃染毒对C57BL/6雄性小鼠免疫功能的影响。方法选择48只SPF级C57BL/6雄性小鼠按体重随机分为4组,分别为对照组(2%Tween-80)和5、20、40 mg/kg PFOS染毒组,每组12只。采用灌胃方式进行染毒,每天染毒1次,连续染毒7 d。利用双抗体夹心ELISA试剂盒检测脾脏淋巴细胞的IL-4和IFN-γ水平;利用ELISpot试剂盒检测脾淋巴细胞中分泌IL-2和IL-10的淋巴细胞数量。结果与对照组相比,5 mg/kg PFOS染毒组小鼠脾脏中分泌IL-2的淋巴细胞数量和脾细胞培养上清中细胞因子IFN-γ水平略微降低,但差异均无统计学意义;但20、40 mg/kg PFOS染毒组小鼠脾脏中分泌IL-2的淋巴细胞数量和脾细胞培养上清中细胞因子IFN-γ均显著低于对照组(P<0.05)。5、20 mg/kg PFOS染毒组小鼠脾细胞培养上清中细胞因子IL-4水平高于对照组(P<0.05);而40 mg/kg PFOS染毒组与对照组差异无统计学意义。各剂量PFOS染毒组小鼠脾脏中分泌IL-10的淋巴细胞数量与对照组相比,差异均无统计学意义。随着PFOS染毒剂量的升高,小鼠脾细胞培养上清中细胞因子IFN-γ水平和脾脏中分泌IL-2的淋巴细胞数量呈下降趋势。结论机体在短期、高剂量PFOS暴露后,处于细胞免疫相对受抑制而体液免疫相对增强的状态。
Objective To investigate the immune state of male C57BU6 mice after high dose and short time PFOS exposure. Methods Forty-eight aduh C57BIJ6 male mice (SPF) were divided into four groups. Animals were given 0,5,20 and 40 mg/kg PFOS by gavage every day for 7 consecutive days, and then single cell suspensions of splenocytes were prepared. After 48 h of culture, the supernatant was collected and was used to check the level of eytokine IL-4 and IFN-γ by ELISA. The number of eytokines IL-2 and IL-10 secreting cells were measured by ELISpot. Results Compared with the control group, the levels of IL-2 and IFN-γ were significant lower in 20 and 40 mg/kg PFOS treated groups (P〈0.05), the levels of IL-4 were higher for 5 and 20 mg/kg PFOS treated group (P〈0.05), but no statistics significance for 40 mg/kg PFOS treated group (P〉0.05); and for IL-10, no significant difference was seen in all the groups (P〉0.05). Conclusion After high dose and short time PFOS exposure,the cellular immunity may be suppressed and the humoral immunity may be enhanced.
