人胎中后期腭扁桃体组织结构特征及其CD20和CD3阳性细胞的表达 被引量:1

摘要 目的从形态学角度探讨人胎中后期腭扁桃体组织结构特征及其CD20和CD3阳性细胞的表达情况。方法收集6~10月龄因故终止妊娠人胎腭扁桃体25例,用组织学常规H-E染色显示其组织结构,免疫组织化学SP法染色显示阳性细胞并用BioMiaspro图象分析软件计数。结果自6月龄起,人胎早期形成的腭扁桃体雏形逐渐完善,固有层增厚,弥散淋巴组织和初级淋巴小结增多,已有大量淋巴细胞表达CD20或CD3。至8月龄起,腭扁桃体隐窝上皮内始见毛细血管浸润。邻片对照显示,CD20阳性细不仅大量分布于淋巴小结,且在弥散淋巴组织和隐窝上皮内也较多;CD3阳性细胞则主要分布于弥散淋巴组织,淋巴小结和隐窝上皮内皆较少。细胞计数显示6~10月龄人胎腭扁桃体,2种阳性细胞数量随胎龄增加逐渐增多(P<0.05),但CD20阳性细胞占绝大多数(P<0.05)。结论 8月龄起,人胎腭扁桃体形态结构基本成熟;人胎腭扁桃体具有潜在的CD20和CD3合成和释放能力,对胎儿免疫功能的建立和健全发挥着重要作用。 To investigate the structural feature of palatine tonsil tissue in the midanaphase human fetus and the expressions of CD20-positive cells as well as CD3-positive cells,25 samples of human fetal palatine tonsil raged from 6-month to 10-month gestational stages were collected.We employed Hematoxylin and Eosin staining to display the structural feature.While CD20-positive cells and CD3-positive cells were observed by Streptavidin-Peroxidase method,then counted with BioMiaspro photographs analysis software.The experiment result showed that the morphological structure of palatine tonsil was firstly formed at 6-month gestational stage.Compared to the earlier period,in 6-month gestational stage,the proper layer was thickened with the increasing of diffused lymphoid tissue and primary lymphoid nodule.At the same time,the expression of CD20 and CD3 were detected on the membrane surface of most lymphocytes.Furthermore,at 8-month gestational stage,blood capillary was firstly seen in the crypt epithelium.The neighboring slice comparison displayed that the distribution of CD20-positive cells was mostly in the primary lymphoid nodule,while CD3-positive cells was mainly in the diffuse lymphoid tissue.Furthermore,many CD20-positive cells also distributed in the diffuse lymphoid tissue and crypt epithelium,but few CD3-positive cells distributed in the primary lymphoid nodule and crypt epithelium.The cell counting displayed that the two types of positive-cells were increased gradually with fetal age growing(P 0.05),and the numbers of CD20-positive cells were majority(P 0.05).This study suggest that the morphological structure of palatine tonsil become elementary mature since 8-month gestational stage,and the palatine tonsil may have potential capacity to synthesize and release CD20 and CD3,thus plays an important role in the establishment and mature of fetal immunological function.
