摘要
核糖核酸是一类十分重要的生物大分子,它在基因的表达和蛋白质的生物合成中均起着关键的作用。以茶树菇为原料,核糖核酸得率为指标,采用响应面分析法优化微波提取工艺,考察微波辐射频率、微波时间、料液比等因素对茶树菇核糖核酸提取率的影响。结果表明,微波辅助提取茶树菇核糖核酸的最佳工艺条件为:微波功率495W,微波时间7min,液料比18∶1,水浴浸提时间2.0h,茶树菇核糖核酸得率可达4.53%。
RNA is a kind of important biological macromolecules,its gene expression and protein biosynthesis plays a key role in absorption.Agrocybe aegeritaw as raw material,the RNA yield as an index,then using response surface method to optimize the process of microwave extraction,the factors,such as ultrasonic power,ultrasonic time,liquid to solid ratio and the water extracting time,which influence the extraction results of the Agrocybe aegeritaw RNA extraction rates were discussed.The results showed that the optimum conditions of microwave-assisted extraction of RNA Agrocybe aegeritaw were:microwave power 495W,ultrasonic time of 7min,liquid feed ratio 18∶1,water bath extraction time 2.0h,Agrocybe aegeritaw RNA yield up to 4.53%.
