低强度复合振动治疗去卵巢老龄大鼠骨质疏松实验研究 被引量:4

摘要 目的:观察低强度复合振动对去卵巢老龄大鼠骨质疏松的治疗作用。方法:13月龄SD雌性经产大鼠16只,随机均分为对照组和实验组。卵巢摘除法骨质疏松造模,3个月后检查骨密度(BMD)下降后实验组接受低强度复合振动(振动频率45 Hz~55 Hz,振动强度0.12 g~0.21 g,g=9.8 m/s2,20分钟/次/天,5天/周),对照组不进行振动治疗。13周复合振动实验结束后分别活体测量两组大鼠腰椎、股骨骨密度(Lorland XR-46),胫骨标本骨微结构扫描分析(SCANCO u-CT),测试腰椎、股骨标本力学性能(858 mini bionix)及酶法检测血清碱性磷酸酶、非前列腺酸性磷酸酶,电化学发光免疫法检测血清骨钙素和I型胶原C端交联肽。结果:复合振动实验结束后实验组腰椎和股骨BMD较实验前显著增加(分别增加0.0110±0.0048 g/cm2,P=0.000和0.0056±0.0065 g/cm2,P=0.004),而对照组显著下降(分别下降0.0071±0.0042 g/cm2和0.0073±0.0063 g/cm2)。相较对照组,骨微结构参数Tb.N显著增加(P=0.002)、Tb.Sp显著降低(P=0.005)以及BV/TV显著增加(P=0.048),腰椎、股骨强度极限也显著增加(P=0.038,P=0.014),但血清骨钙素、碱性磷酸酶、I型胶原C端交联肽、非前列腺酸性磷酸酶两组无统计学差异。结论:低强度复合振动可以减轻去卵巢老龄大鼠骨质疏松程度,提高骨质疏松大鼠骨质量。 Objective To observe the effect of low-magnitude compound vibration to ovariectomized old SD rat' s osteoporosis.Methods Sixteen 13-months-old multiparous SD rats were randomly divided into 2 groups: control(n = 8) and vibration group(n = 8).All the rats were ovariectomized to model osteoporosis for 3 months.The experiment of low-magnitude compound vibration began when bone mineral density(BMD)decreased.The vibration group received low-magnitude compound vibration(vibration frequency:45 Hz~55 Hz,vibration magnitude:0.12 g ~ 0.21 g,g = 9.8 m/s2,20 mins/times·day,5 days/week) and the control group kept sedentary.The parameters related to osteoporosis of the two groups rats were measured after the 13 weeks compound vibration trial,such as:the intravital lumbar and femur BMD(Lorland XR-46),bone microarchitecture of tibia specimen(SCANCO u-CT),biomechanical property of lumbar vertebra and femur(858mini bionix),blood serum alkali phosphatase and acid phosphatese of non-prostate(enzymic method),blood serum osteocalcin and carboxyterminal crosslinked telopeptide of typeⅠcollagen(electrochemiluminescence immunization).Results Lumbar and femur BMD of vibration group rats increased significantly(0.0110±0.0048 g/cm2,P =0.000 and 0.0056±0.0065 g/cm2,P = 0.004,respectively) at post-vibration compared with pre-vibration,while the control group decreased accordingly(-0.0071±0.0042 g/cm2 and-0.0073±0.0063 g/cm2).Significant improvement were found on some parameters of bone microstructure and bone biomechanics,such as Tb.N(P = 0.002),Tb.Sp(P = 0.005),BV/TV(P = 0.048) and bone ultimate strength(P = 0.038,0.014).But there was no statistics difference on the bone metabolism markers between the two groups.Conclusion Low-magnitude compound vibration could slow the process of osteoporosis and enhance the bone quality of ovariectomized old rats.
