黄芪皂甙、丹参酮ⅡA注射液对大鼠骨骼肌急性钝挫伤后骨骼肌组织形态的影响

Effects of Astragaloside and Tanshinone ⅡA Injection on the Skeletal Muscle Morphology in Rats with Acute Muscle Contusion

目的:观察黄芪皂甙、丹参酮ⅡA注射液对大鼠骨骼肌钝挫伤后骨骼肌组织形态的影响。方法:84只雄性Wistar大鼠随机分为6组,包括5个干预组(每组16只)和1个正常对照组(4只)。用打击装置将干预组大鼠双侧腓肠肌中段造成钝挫伤模型,损伤即刻在局部分别注射黄芪皂甙、丹参酮ⅡA、黄芪皂甙丹参酮ⅡA混合液、黄芪丹参混合液及生理盐水。以后每3天注射1次,直到损伤后第27天。分别于损伤后第4天、7天、14天和28天从双侧腓肠肌损伤部位取材,每组4只。正常对照组于第28天进行双侧腓肠肌取材。经HE染色观察骨骼肌纤维直径(横断面),经Masson染色观察损伤处纤维疤痕组织面积(纵切面)。结果:(1)各组不同时间点骨骼肌纤维直径无统计学差异。(2)各药物干预组纤维疤痕面积均显著小于生理盐水组,黄芪皂甙丹参酮ⅡA混合组纤维疤痕面积显著小于黄芪皂甙组和丹参酮ⅡA组,黄芪皂甙丹参酮ⅡA混合组与黄芪丹参组结果无统计学差异。结论:在急性骨骼肌钝挫伤修复过程中,黄芪皂甙、丹参酮ⅡA注射液能减少损伤区域纤维疤痕组织形成,对损伤部位骨骼肌纤维直径无明显影响。

Objective To study the effects of astragaloside and tanshinone ⅡA injection on the skeletal muscle morphology during the repair process after skeletal muscle contusion.Methods Eighty four male Wistar rats were divided to six groups：five intervention groups（16 rats in each group）：astragaloside,tanshinone ⅡA,mixed injection（astragaloside and tanshinone ⅡA）,Huangqi-Danshen（Milkvetch Root-Danshen Root） injection and physiological saline groups,and a normal control group（4 rats in this group）.In intervention groups,bilateral gastrocnemius of rats were contused by specific device,then the contused muscles were injected with the above corresponding materials.Those injections were executed every 3 days until 27 days after injury.Samples were collected from injured muscles 4,7,14 and 28 days after the injury.In the normal control group,the bilateral gastrocnemius were collected in the 28th day.The diameter of skeletal muscle fiber（cross section） in HE stain and the area of fibrous scar tissue（longitudinal section）in Masson stain were observed.Results（1）In all groups,the diameters of skeletal muscle fiber had no significant difference at the different time courses.（2）The area of fibrous scar in the intervention groups was smaller than in the saline group,and the area of fibrous scar in the mixed injection group was smaller than in astragaloside group and tanshinone ⅡA group,while there was no difference between the mixed injection group and the Huangqi-Danshen group.Conclusion During the repair process following acute contusion of skeletal muscle,astragaloside and tanshinone ⅡA injection can reduce the area of fibrous scar tissue without any effect on the diameters of skeletal muscle fiber.
