肝细胞生长因子对大鼠背部超长任意皮瓣存活率影响的实验研究 被引量:3

摘要 目的:探讨肝细胞生长因子对大鼠背部超长任意皮瓣血管生成的作用及皮瓣存活率的影响。方法:建立大鼠背部超长任意皮瓣8.0cm×2.0cm动物模型,直接皮下注射50ng/ml肝细胞生长因子溶液,术后7天观察皮瓣颜色、质地、毛细血管回流、坏死范围、切割皮瓣出血情况;计算皮瓣的存活面积;免疫组化染色后在显微镜下计算毛细血管平均密度。通过自身前后对照、与生理盐水组对照,比较两者的差异。结果:肝细胞生长因子组皮瓣的存活率81.45%±2.74%,与对照组的42.82%±7.03%比较,差别有统计学意义(P<0.05);实验前实验组和对照组毛细血管数分别为12.70±1.35、12.31±1.22,两者差别无统计学意义(P>0.05);实验后实验组和对照组毛细血管数分别为39.67±4.83、21.50±1.87,两者差别有统计学意义(P<0.05);实验组及对照组实验后毛细血管密度高于实验前,差别有统计学意义(P<0.05)。结论:肝细胞生长因子能促进皮瓣血管内皮细胞的生成,提高皮瓣的存活率。 Objective To investigate the angiogenesis function of hepatocyte growth factor(HGF) to random ultra-long skin flap on the rat back and its effect on the survival rate of the skin flap.Methods Making a 8.0cm×2.0cm animal model from a random ultra-long skin flap on the rat back,injected directly with 50ng/ml HGF solution,after 7 days observing the color of the skin flap,texture,capillary return,necrotic area,bleeding condition of the cut flap etc.;Calculating the survival area of the flap;After immunity staining,counting blood capillary with a 400 optical microscope.Making results through comparing the disparity between the before-experimental flap and that of after——experiment.,And also comparing with the saline control group.Results Comparing the survival rate(81.45%±2.74%) of the HGF skin flap with the control group(42.82%±7.03%),the differences are statistically significant(P 0.05);before experiment,The quantity of capillary of the experimental group and the control group are respectively(12.70±1.35) and(12.31±1.22),the difference between the two is of statistic significance(P0.05);After experiment,the quantity of capillary of the experimental group and the control group are respectively(39.67±4.83) and(21.50±1.87),The difference between the two is of statistic significance(P0.05);After experiment,the number of capillary of the experimental group is higher than that before experiment,the difference is statistically significant(P0.05).Conclusion HGF can promote the formation of the vascular endothelial cells in the flap and improve the survival rate of skin flaps.
