摘要 转录因子AP-2是一个功能重要的基因家族,AP-2α在协同调控脂肪细胞分化成熟,以及脂肪因子分泌的过程中发挥重要作用。为了阐明其协同调控的机制,本文采用anti-AP-2α抗体免疫共沉淀技术,从小鼠前脂肪细胞3T3-L1中分离AP-2α相互作用蛋白,然后用基质辅助激光解吸/电离飞行时间质谱结合数据库查询鉴定AP-2α结合蛋白,找到了acyl-Coenzyme A dehydrogenase,long-chain、Protein-L-isoaspartate(D-aspartate)O-meth-yltransferase和T-complex proteom 1 subunit epsilon三个与AP-2α相互作用的蛋白。为进一步研究小鼠的前脂肪细胞中AP-2α蛋白聚集及在糖脂代谢调控中可能的作用机制提供了重要依据。 Transcription factor AP-2 is an important gene family, AP-2α plays a major role in the coordination regulation of adipoeyte differentiation, maturation and adipoeyte factor secretion. To elucidate the mechanism of the coordination regulation, this paper carries out the co-immunoprecipitation method using the anti-AP-2α antibodies, ap-2-interacting proteins were isolated from mouse pre-adipocyte cells, these binding factors were identified by MALDI-TOF-MS and database search, they are acyl-Coen,'~,yme A dehydrogenase, long-chain, Protein-L-isoaspartate (D-aspartate) O-methyl- transferase and T-complex protein 1 subunit epsilon, which provides a significant basis for the further research of AP-2α protein accumulation in mouse pre-adipocyte and the regulation mechanism of glycolipid metabolism.
