基质固相分散提取桃儿七根中鬼臼毒苷和异鬼臼苦酮 被引量:8

Matrix Solid Phase Dispersion for the Extraction of Podophyllotoxin Glucoside and Isopicropodophyllone from Roots of Sinopodophyllum emodi(Wall.)

摘要 利用基质固相分散法(MSPO)提取桃儿七根中的鬼臼毒苷和异鬼臼苦酮,并采用高效液相色谱法对提取结果进行了分析.结果表明,对于鬼臼毒苷,以硅胶作为分散剂,以甲醇或乙醇作为洗脱剂时其提取率最高;对于异鬼臼苦酮,以硅藻土作为分散剂,以甲醇或乙醇作为洗脱剂时其提取率最高.本方法对鬼臼毒苷和异鬼臼苦酮的提取率分别为18.40和0.60 mg/g,高于传统的甲醇回流提取(分别为17.55和0.38 mg/g),尤其对异鬼臼苦酮的提取率明显高于甲醇回流提取. A new extraction procedure based on the matrix solid-phase dispersion(MSPD) was proposed for the extraction of two kinds of lignans including podophyllotoxin glucoside and isopicropodophyllone from the dried roots of Sinopodophyllum emodi(Wall.) Ying.The compounds were determined by high performance li-quid chromatography.According to the results,the podophyllotoxin glucoside got high extraction yield when silica gel was applied as dispersant and methanol/ethanol was used as elution solvent.For isopicropodophyllone high extration yield was got when diatomaceous earth was used as dispersant and methanol was used as elution solvent.The extraction yields by MSPD method were 18.40 and 0.60 mg/g,respectively.Compared with methanol reflex extraction,the values were only 17.55 and 0.38 mg/g.In conclusion,MSPD method is more appropriate for the extraction of the two lignans than the methanol reflux method,especially for the extraction of isopicropodophyllone.Moreover,MSPD method is fast,simple,high-yield with less solvent.
