摘要
目的研究建立菊淀粉型寡糖的高效液相色谱(HPLC)测定方法,定量测定巴戟天中菊淀粉型四、五、六、七寡糖的含量。方法提取巴戟天寡糖,直接分析。色谱柱:Click Maltose 4.6 I.D.×100 mm,流动相:乙腈-水,流速1.0 ml/min,100 ELSD9型蒸发光散射检测器(ELSD),进样量20μl,用外标法测定巴戟天寡糖中四~七糖的含量。结果建立了测定巴戟天寡糖中四~七糖含量的方法。四~七糖的线性范围分别为四糖11~90.72μg,r=0.9996(RSD=2.2%);五糖5.67~90.72μg,r=0.9992(RSD=1.56%);六糖9.6~48.78μg,r=0.9998(RSD=3.2%);七糖6.8~33.6μg,r=0.9984(RSD=2.7%)。结论本方法可以作为新药巴戟天寡糖的质量控制方法,也可用于巴戟天药材的质量控制。
Objective To establish a method,using HPLC,for determination of 4-7 sugars in oligosaccharides of Morinda officinalis How..Method Oligosaccharides of M.officinalis How.were extracted by routine method and used for the HPLC analysis directly.The samples were subjected to chromatography on an Inertsil Click Maltose 4.6 I.D.×100 mm column and eluted with acetonitrile-H2O at a flow rate of 1 ml/min.The samples were determined with a 100-ELSD9 ELSD detector and calibrated using the external standards method.The contrast sample was used with 4-7 oligosaccharides respectively.Result A determination method of the 4-7 sugars in oligosaccharides of M.officinalis How.was established.Satisfactory linearity and accuracy of the 4-7 sugars were obtained,with a range of 11-90.72 μg(r=0.9996,RSD=2.2% 4 sugar);5.67-90.72 μg(r=0.9992,RSD=1.56%,5 sugar);9.6-48.78 μg(r=0.9998,RSD=3.2%,6 sugar);and 6.8-33.6 μg(r=0.9984,RSD=2.7%),respectively.Conclusion The method can be used for the determination of 4-7 sugars of oligosaccharides of M.officinalis How.of a new drug or as a reference of other kind oligosaccharides.
