化学刺激致溃疡性结肠炎兔模型的建立

Chemical stimulation induced ulcerative colitis in rabbit models

目的:建立溃疡性结肠炎兔模型并探讨其应用价值。方法:30只成年健康新西兰兔随机分为5组,每组6只,实验组分别用5%乙酸、8%乙酸、10%乙酸灌肠造模,阳性对照组以三硝基苯磺酸(TN-BS)灌肠造模,空白对照组以生理盐水灌肠,比较各组疾病活动指数(DAI),并进行组织学观察。结果:5%乙酸造模组DAI与TNBS对照组无统计学差异(P>0.05),10%乙酸造模组与其他各组差异均有统计学意义(P<0.05～0.01)。组织学观察:除空白对照组外,各组均发现肠壁增厚,黏膜弥漫性出血、溃疡、坏死等情况,其中5%乙酸造膜组黏膜炎症细胞浸润,腺体消失,而另外3个模型组黏膜均严重坏死脱落。结论:5%乙酸局部刺激能较好复制兔溃疡性结肠炎模型,该模型制作方法简单,成本低,周期短,成功率高,重复性好,是较为理想的溃疡性结肠炎动物模型。

Objective：To establish a rabbit model of ulcerative colitis.Methods：Thirty adult white rabbits of New Zealand were divided into five groups randomly,each having 6 animals.The experimental groups were treated with 5%,8% and 10% acetic acid to induce the enema modeling.TNBS enema modeling（positive control group） and negative control group were also created for comparison of DAI among these groups.Historical analysis was also performed.Results：The group treated with 5% acetic acid showed no significant difference in DAI comparing with the positive control group（P0.05）,but the group treated with 10% acetic acid group showed significant differences when comparing with the rest 4 groups（P0.05～0.01）.Histological observation revealed that increased intestinal walls,mucosal bleeding,ulcers,and necrosis.Specifically,the 5% acetic acid treated group showed inflammatory,cell infiltration and disappearance of glands,etc.The other 3 groups also showed mucosal necrosis and fall.Conclusions：Topical stimulation of 5% acetic acid can induce ulcer colitis in rabbit model.It＇s also cheap,quick and has good reproducibility.
