双酚A对人早孕绒毛组织IGF1、TGFβ1、ADAM12表达的影响 被引量:4

摘要 目的:研究双酚A(BPA)水平与人早孕胰岛素样生长因子1(IGF1),转化生长因子β1(TGFβ1),以及解整合素样金属蛋白酶12(ADAM12)表达的关系,探讨BPA对绒毛组织可能带来的影响。方法:以40例早期妊娠(6~8)周的新鲜绒毛组织进行体外培养,培养基中加入不同浓度的BPA:A组为1 000×10-6g/L,B组为500×10-6g/L,C组为1×10-6g/L,D组为对照组。收集培养第1天,培养第3天的培养液,采用ELISA方法检测收集的培养上清中TGFβ1、ADAM12及IGF1的分泌水平。结果:绒毛组织培养液第3天的TGFβ1、ADAM12的浓度降低(P<0.05)。第3天培养液中IGF1浓度变化不大(P>0.05)。结论:BPA对绒毛组织具有细胞毒性作用。 Objective:To study the effects of bisphenol A(BPA) on the expression of IGF1,TGFβ1,and ADAM12 in chorionic villus early in pregnancy.Methods:Villus samples collected from 40 early pregnancy cases(6~8 weeks) were cultured(in vivo) and treated with BPA at different concentrations.The concentrations of BPA was 1 000×10-6g/L for group A,500×10-6g/L for group B,1×10-6g/L for group C,respectively,group D was untreated control group.On the 1st,3rd day,IGF1、TGFβ1、ADAM12 concentrations of supernatant in the culture were measured by ELISA.Results:The concentrations of TGFβ1,ADAM12 were significantly decreased on day 3(P0.05),but IGF1 showed little changes in concentrations(P0.05).Conclusion:BPA is cytotoxic to villus during early pregancy.
