摘要
目的探讨七叶亭(Esculetin)预处理对大鼠急性心肌缺血再灌注损伤(MIRI)的保护作用。方法将48只雄性SD大鼠随机分为假手术组(Sham组)、缺血再灌注损伤组(MIRI)、七叶亭预处理组(esculetin组)、缺血预处理组(IPC组)。测定血清肌酸激酶(CK)、肿瘤坏死因子(TNF-α)、IL-10水平,测量心肌梗死范围,Westernblot法检测NF-κB。结果与Sham组比较,其余各组血清TNF-α、IL-10、CK水平明显增高(P<0.05),梗死范围明显增加(P<0.05),且NF-κB表达明显增高(P<0.05)。esculetin组、IPC组TNF-α、IL-10、CK水平无统计学差异;两组间心肌梗死的范围、NF-κB蛋白水平无统计学差异。结论七叶亭通过抑制炎症而发挥对缺血再灌注心肌的保护作用。
Objective To study the protective effect of esculetin on acute myocardial ischemia reperfusion in rats.Methods Forty eight SD rats were randomly assigned into four different groups,ie.sham group,ischemia reperfusion injury(MIRI)group,esculetin preconditioning(esculetin)group and ischemia preconditioning(IPC)group.The levels of serum creatine kinase(CK),tumor necrosis factor-alpha(TNF-α)and interleukin-10(IL-10)were detected.The infarct size of each group were measured by TTC staining.Western blot was used to monitor the levels of NF-κB.Results Compared with shame group,serum concentration of TNF-α,IL-10 and CK,infarct size and the expression of NF-κB significantly increased in MIRI group,esculetin group and IPC group(P0.05).The levels of TNF-α,IL-10,CK and NF-κB as well as infarct size had no significantly statistical difference in esculetin group and IPC group.Conclusion Esculetin can protect myocardial from ischemia reperfusion injury by reducing systemic inflammatory responses.
