摘要
考察了3,4-二氨基苯基苯甲酮(DABP)作为基质辅助激光解吸离子化飞行时间质谱(MALDI-TOF-MS)新型基质分析磷脂的能力。与传统基质2,4,6-三羟基苯乙酮(THAP)、2,5-二羟基苯甲酸(DHB)、α-氰基-4-羟基肉桂酸(CHCA)相比,用优化后的DABP基质溶液体系MeOH-H2O-HCl(80∶20∶3,V/V)分析不同种类的磷脂,可以有效抑制碱金属离子加成现象,得到单一的质子加成峰。样品信号强,所需激光能量低,卵磷脂(PC)的检出限可达到500 fmol。在实际样品鸡蛋和大豆提取物分析过程中,DABP基质对磷脂分子具有良好的分辨率,碎片峰少,谱图背景干净,样品制备方法简单省时。
The matrix of 3,4-diaminobenzophenone(DABP) was applied in the analysis of phospho-lipids by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.The optimal composition of DABP matrix solution with methanol-water-hydrochloric acid(80 ∶ 20 ∶ 3,V/V) was used in the analysis of different phospholipids species by comparing with the conventional matrices such as 2,4,6-trihydroxyacetophenone(THAP),2,5-dihydroxy-benzoic acid(DHB) and α-cynao-4-hydroxycinnamic acid(CHCA),3,4-diamino-benzophenone(DABP) can effectively suppress alkali metal ion adducts,thus showing single peak of phospholipids with low laser energy and strong peak strength.500 fmol phosphatidylcholines(PCs) can be detected with high signal to noise ratio.With DABP as a matrix for the analysis of egg yolk and soybean extracts,intact phospholipids ion peaks can be readily produced with clean spectrum background and high resolution.In addition,the sample preparation method is simple and time-saving.
