高钼对雏鸡肝细胞周期和凋亡的影响 被引量:6

Effect of dietary high molybdenum on the cell cycle and apoptosis of liver in broilers

摘要 300只1日龄健康Avian肉鸡随机分为4组,分别喂以对照组日粮(Mo 13 mg/kg)和高钼日粮(Mo 500 mg/kg,高钼Ⅰ组;Mo 1 000 mg/kg,高钼Ⅱ组;Mo 1 500 mg/kg,高钼Ⅲ组)6周,于14,28,42日龄每天随机抽取5只以流式细胞术研究高钼对雏鸡肝脏细胞周期和凋亡的影响。结果显示:高钼Ⅱ组和Ⅲ组雏鸡体质量和肝脏质量显著低于对照组(P<0.01或P<0.05),脏器指数显著高于对照组(P<0.01或P<0.05)。电镜下肝细胞内质网扩张,线粒体肿胀、空泡化。流式细胞仪测定,高钼Ⅱ组和Ⅲ组肝细胞G0/G1期极显著升高(P<0.01),S期、G2+M期和增殖指数(PI)与对照组相比不同程度地降低;同时,高钼Ⅱ组和Ⅲ组肝细胞凋亡率均极显著高于对照组(P<0.01)。结果表明,日粮含钼1 000 mg/kg及以上时可引起肝细胞分裂增殖能力受阻,凋亡细胞显著增多。 Three hundred 1-day-old Avian broilers were divided into four groups and fed on control diet(Mo 13 mg/kg) and high molybdenum diets(Mo 500 mg/kg,groupⅠ;Mo 1 000 mg/kg,groupⅡ;Mo 1 500 mg/kg,group Ⅲ) for 6 weeks.The effect of dietary high molybdenum on the cell cycle and apoptosis of liver in chickens were researched by the methods of flow cytometry(FCM) at 14,28,42 d old.The weight of body and liver was significantly decreased(P0.05 or P0.01),and the relative weight of liver was significantly increased in group Ⅱand Ⅲ compared with control group(P0.05 or P0.01).Ultrastructurally,endoplasmic reticulums of hepatocyte were dilated and the mitochondria were swelled or with its crista broken.By FCM,the G0/G1 phase was significantly increased(P0.01),and S phase,G2+M phase and PI(proliferating index) significantly decreased(P0.01) in groups Ⅱ and Ⅲ during this experiment.The apoptotic rate was higher in group Ⅱ and Ⅲ than that in control group at 14,28 and 42 days of age(P0.01).The result showed that dietary molybdenum in excess of 1 000 mg/kg diet could result in the restrained multiplication of hepatocyte and increased apoptosis.
