摘要
目的:研究康脑液2号对脑缺血再灌注大鼠血管内皮生长因子(VEGF)、血管生成素-1(Ang-1)与微血管密度(MVD)的影响。方法:180只雄性SD大鼠随机均分为正常组、假手术组、模型组与康脑液2号高、中、低剂量组。线栓法复制大鼠右侧大脑中动脉闭塞模型。大鼠缺血2h,于再灌注1、3、7、14、21d,免疫组化法检测各组大鼠VEGF、Ang-1的表达,HE染色观察大鼠脑组织病理改变;再灌注21d后免疫荧光法标记细胞膜糖蛋白CD105,观察新生血管,计算MVD。结果:与模型组比较,康脑液2号高、中、低剂量组病理变化更显著,VEGF、Ang-1在每个时间点表达显著增高,脑梗死周围区域MVD显著增大。结论:康脑液2号能改善脑缺血再灌注大鼠病理变化,保护梗死区域神经细胞,其机制可能与增加VEGF、Ang-1等促血管新生因子表达而促进脑坏死周围区域血管新生有关。
OBJECTIVE:To investigate the influences of Kangnao liquid Ⅱ on vascular endothelial growth factor(VEGF) ,angiogenin-1(Ang-1) and MVD in rats with focal cerebral ischemia reperfusion injury. METHODS:180 healthy male SD rats were randomly divided into normal group,sham operation group,model group,Kangnao liquid Ⅱ high-dose,medium-dose and low-dose groups. Middle cerebral artery occlusion model was induced using suture method. Pathological changes were detected by HE staining and immunohistochemical method was used to investigate the expression of VEGF and Ang-1 in rats on 1 d,3 d,7 d,14 d,21 d of reperfusion injury after ischemia for 2 h. Immunofluorescence labeling method was used to mark neovascular CD105 after 21 d of reperfusion and calculate MVD. RESULTS:Compared with model group,pathological changes of Kangnao liquid Ⅱ high-dose,medium-dose,low-dose groups was better than that of model group. The expressions of VEGF and Ang-1 increased at different time points significantly. MVD of cerebral infarction area was significant bigger than that of model group. CONCLUSION:Kangnao liquid Ⅱ can improve pathological change of rats with cerebral ischemia-reperfusion injury and protect nerve cells of cerebral infarction focus. It may be associated with Kangnao liquid Ⅱ can increase the expression of VEGF and Ang-1 to promote tissue angiogenesis.
